What Are The Core Benefits And Application Areas Of Neutral Protease?

what is Neutral Protease?
Neutral proteases are metalloproteinases that function under neutral pH conditions (optimal pH 6.0–8.0) and are primarily derived from Bacillus polymyxa.
Their core active site consists of zinc ions and a conserved amino acid sequence (histidine-glutamic acid-histidine). They specifically hydrolyze fibronectin, laminin, and type IV collagen in the extracellular matrix, but exhibit low activity in degrading fibrous collagen.
neutral protease benefits:
1. Mildness and Efficiency
- It operates efficiently at physiological pH (7.0–8.0) and temperature (37°C), avoiding cell damage from strong acid/alkali environments.
- It precisely degrades cell-matrix junction proteins, preserving cell surface receptors and intercellular junction structures, significantly improving cell activity and functional integrity.
2. High Stability
Neutral protease enzyme is insensitive to serum components, and its stability can be enhanced by adding Ca²⁺; chelating agents such as EDTA can rapidly terminate enzyme activity, facilitating reaction control.
3. Good Safety
Produced by bacterial fermentation, it carries no risk of mycoplasma or animal virus contamination, making it suitable for clinical-grade cell preparation.
neutral protease uses:
1. Life Sciences and Medical Research
- Primary Cell Isolation: Isolation of neural stem cells, extraction of epithelial cells
- Organoids and Cell Passaging
- Tissue Engineering: Isolation of intact epidermal and dermal layers, or release of epithelial lamellae from culture media for skin regeneration research.
2. Food Industry
- Baking Industry: Hydrolyzes dough proteins, reduces gluten strength, improves biscuit plasticity and texture, and promotes Maillard reactions to enhance flavor.
- Beer Clarification: Degrades cloudy proteins; when combined with other enzymes, it can remove bitterness.
- Protein Hydrolysate Production: Prepares hydrolyzed animal and plant proteins for use in flavorings and nutritional fortifiers.
3. Feed Additives
Neutral protease powder can improve the utilization rate of aquatic feed, promote nutrient metabolism and weight gain in fish.
